Production, Purification, Mutagenesis and Characterization of proteins expressed in E. coli

In recent years, the research activity has concerned, mainly and not without difficulty but with great enthusiasm and passion, the production, purification and mutagenesis of proteins expressed in E.coli. We produce variants of neuroserpin (NS), both pathological and non-pathological, and we study the effects of these mutations, in vitro and in silico, to understand the neuroserpin polymer formation mechanisms (NS) and to obtain information on the structure and molecular flexibility.
The activity includes:
- production of wild-type and site-point mutations of NS and subsequent EPR and Fluorescence
measurements;
- in silico study of cavity identification on the molecular surface of NS for the research of small
molecules that inhibit polymerization;
- use of new eukaryote expression systems for the production of serpines;
- study of the interaction of neuroserpine with neuronal proteins.
This research activity includes the project financed by the European Social Fund - Sicily Region (Bando N. IBF-BS-001-2018-PA /SPIN OFF), with the main objective of favoring the working orientation of young people towards the research and innovation system.
