Quantitative single molecule localizations of channel proteins by using fluorescence super resolution tecniques

Far field super-resolution microscopy techniques based on single molecule localization STORM and dSTORM is a novel tool to quantify protein distribution and to “count” molecules in biological systems. Single molecule approaches, based on stepwise photobleaching, have also been developed to meet the need to characterize subunits of protein homo- and hetero-multimers, thus allowing quantitative characterization and localization of membrane channels Furthermore, efforts have been recently focused on the application of quantitative approaches, based on clustering analysis and molecular counting, in order to address specific biological questions.
